Labeling MCF-7 Cell Line with Interleukin-6/Calcium Phosphate Nanoparticles

In recent years, nanoparticle systems have been frequently used as non-viral carriers for the treatment of genetically based diseases and cancer. However, conjugate formation should be optimized by selecting a suitable carrier transport each genetic material desired to transported target cell. A created taking into account size zeta potential electrical characteristics cargo being transported. Nanocarrier can protect package material. Calcium phosphate nanoparticles (CaP NPs) with unique advantages promising approach in vitro and in vivo gene transfers. biggest challenge CaP NPs is their unreducible size. this study, were synthesized via co-precipitation method using two stabilizing agents separately: magnesium nitrate sodium citrate. The transfection efficiency obtained citrate MCF-7 cell line was observed more effective compared nitrate-containing nanoparticles. (248.1 ± 54) nm (polydispersity index (PDI), 0.3) (–13.3 2.8) mV synthesized. morphological properties particles determined scanning electron microscopy (SEM) atomic force (AFM). Cytotoxicity performed 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) analysis L929 mouse fibroblast cells. both CaP/plasmid DNA conjugation naked plasmid transferred cells, expression fluorescent green protein (GFP) expression. Gene efficiencies 24% (±1.7%) pDNA 76% (±2.3%) CaP-conjugated pDNA, respectively.
